In vitro Growth and Nutrient Uptake of Campbell Early Grapes as Influenced by Some Modifications of MS Media



Abstract
Shoot tips derived from the cuttings of Campbell Early (L¢¥itis labruscana B.) grapes were cultured on Murashige and Skoog (MS) media differing in pH, nitrogen level and source (NO©ý-N or NH©þ-N). Cl^-, and SO©þ^(2-) ions, and their in vitro growth and nutrient uptake were investigated.
1. Optimum pH for shoot growth ranged from pH 4.0 to 5.0, and increase in fresh weight was greatest with pH 5.0.
2. Rapid shoot proliferation was obtained with the 0.75-fold nitrogen (0.75N) of the MS medium, and the optimal range for shoot growth was 1.0N to 0.75N. Adequate balance of shoot mineral content was obtained with 0.75N while Ca uptake was greatest with 0.50N.
3. Adequate ratio of NO©ý-N to NH©þ-N for the growth of shoot tips in vitro ranged from 75 : 25 to 50 : 50. The seventy five NO©ý- N: twenty five NH©þ-N medium increased the shoot growth while the fresh weight increase was greater in 100 : 0 medium.
4. MS medium with both Cl^- and SO©þ^(2-) ions was better for shoot growth and nutrient uptake than those lacking in either one of the ions. Medium composed only of SO©þ^(2-) ions gave a greate: increase in fresh weight than other media.
